چکیده

Introduction One essential requirement for solid tumor growth is the ability to acquire an adequate blood supply; therefore, tumor vasculature is in a perpetual state of angiogenesis. Integrin αvβ3 is a well-established biomarker for angiogenesis with overexpression in tumor vasculature and low to no expression in resting endothelial cells. Non-invasive assessment of tumor angiogenesis is important for early diagnosis of cancer as well as post-therapy assessment. 3 Here we describe the use of αvβ3 targeted magnetofluorescent micelles to analyze angiogenesis in tumor xenografts and nanoprobe accumulation and kinetics by 3D magnetic resonance imaging (MRI) and T2*-weighted dynamic contrast enhancement MRI (DCE-MRI).

β3-specific Dual-modality Micellar Nanoprobes

Multimodality tumor-specific nanoprobes that incorporate two or more imaging functionalities are becoming an integral part of early detection, molecular diagnosis and post-therapy assessment of cancer (1,2). Magnetic resonance imaging (MRI) and optical imaging are two complementary imaging modalities that provide superb anatomical and functional imaging information, respectively. Here we report...

In vivo angiogenesis imaging of solid tumors by avb3-targeted, dual-modality micellar nanoprobes

The objective of this study was to develop and evaluate an avb3-specific nanoprobe consisting of fluorescent superparamagnetic polymeric micelles (FSPPM) for in vivo imaging of tumor angiogenesis. Spherical micelles were produced using poly(ethylene glycol)-b-poly(D,L-lactide) co-polymers conjugated with tetramethylrhodamine, a fluorescent dye, and loaded with superparamagnetic iron oxide nanop...

Anti-αvβ3 antibody guided three-step pretargeting approach using magnetoliposomes for molecular magnetic resonance imaging of breast cancer angiogenesis

PURPOSE Pretargeting of biomarkers with nanoparticles in molecular imaging is promising to improve diagnostic specificity and realize signal amplification, but data regarding its targeting potential in magnetic resonance (MR) imaging are limited. The purpose of this study was to evaluate the tumor angiogenesis targeting efficacy of the anti-αvβ3 antibody guided three-step pretargeting approach ...

Anti-angiogenesis therapy in the Vx2 rabbit cancer model with a lipase-cleavable Sn 2 taxane phospholipid prodrug using αvÎ23-targeted theranostic nanoparticles

In nanomedicine, the hydrophobic nature of paclitaxel has favored its incorporation into many nanoparticle formulations for anti-cancer chemotherapy. At lower doses taxanes are reported to elicit anti-angiogenic responses. In the present study, the facile synthesis, development and characterization of a new lipase-labile docetaxel prodrug is reported and shown to be an effective anti-angiogenic...

Fast Spectroscopic Imaging (F-uTSI) of Angiogenesis on Vx-2 Tumors in Rabbits Using Targeted Perfluorocarbon Emulsions

Quantitative molecular MR imaging of angiogenesis may fulfill an unmet clinical need for patient stratification by increasing efficacy of antiangiogenic therapy. In particular, perfluorocarbons targeted to αvβ3 have been used to image angiogenesis through paramagnetic markers and also direct detection by F MR imaging or spectroscopy [1,2,3]. F offers several advantages including absolute quanti...
